CLIA Reagents of Inflammation (Anti-TP)
The kit has been designed for the qualitative determination of Treponema Pallidum Antibody (Anti-TP) in human serum.
The test has to be performed on the CIA fully auto analyzer (Including CIA 600, CIA 1200, CIA 1800).
TP is slender, 5-15 × 0.1-0.2um, like the shape of the fine spring.Spiral bending is in rule, 8-14 pcs on average, with both ends straight. TP structure is very complex under electron microscope, which isdivided into the outer membrane (mainly consisting of proteins, sugars and lipids), axial filament (mainly composed of protein), a cylindrical cell (including the cell wall, cell membrane and cytoplasmic contents) from outside to inside. Is is no easy coloring. TP’s life cycle is divided into particulate period, spheroids period and spirochetes period. Andit proliferates a generation in around 30 hours. Growth cycle relates with the disease-cycle, insidious onset and a chronic course.
TP antigen divides into three categories:
1. Spirochete surface specific antigen: stimulate the body to produce specific agglutination antibodies and Treponema toimmobilize or dissolve the antibody, while added with soluble complement, the latter can dissolve spirochetes.
2. Spiral generic antigens in vivo: produce complement to bind antibodies, and can cross-reacted with non-pathogenic spirochetes.
3, Complex antigen formed by spirochetes and host tissue phospholipid: When spirochetes invade into tissue, tissue phospholipids adhere to spirochetes and form a complex antigen.Such complex antigens can stimulate the antiphospholipid autoimmune antibodies, which is called reagin (Aegagin), can make precipitation reaction (Conradson test) or complement binding reaction (F test)with lipid antigens extracted from the bovine or other normal animal’s cardiac musle.
In 1981, Fieldsteel succeeded to culture the TP on the monolayerepithelial cell of the cottontail rabbit under microaerobic conditions, which is yet not clear on artificial media. Treponema Pallidum is particularly sensitive to temperature and drying, while it will be dead in in vitro drying for 1-2 hours and at 41 ℃ for 1 hour. It is also sensitive to chemical disinfectants, penicillin, tetracycline and arsenic while it will be dead in 1-2% phenol death for a few minutes.
Treponema pallidum is the causative agent of syphilis. It is characterized by its transparency, no easy coloring. Syphilis is a widespread sexually transmitted diseases, and the morbidity in China has rebounded. Treponema pallidum only infects humans, which is divided into syphilis and congenital syphilis. Syphilis in mainly infected through sexual contact; Congenital syphilis is infected through the placenta and can cause fetal systemic infection. Spirochetes multiply in internal organs and tissues of the fetus, which can cause fetal death or miscarriage.
Syphilis is a kind of disease unique to humans.Dominant and recessive syphilis patientsare the source of infection.Lesions, secretions, and blood of infected people contain Treponema pallidum. It is the most infectious in the first 2 years after infection, while less infectious through sexually transmission after four years of infection. Treponema pallidum can be transmitted to the fetus through the placenta, so it is of great danger to the fetus for early syphilis-infected pregnant women.
